The most reliable individual markers of biotin status, including deficiency and sufficiency, are biotinylated MCC and propionyl-CoA carboxylase in white blood cells [ 7 ]. Oral administration of large doses of biotin increases serum concentrations of biotin and its metabolites [ 1 , 9 ].

However, serum concentrations of biotin and its catabolites are not good indicators of marginal biotin deficiency because they do not decrease sufficiently in people with marginal biotin deficiency for these changes to be detectable with existing tests [ 3 , 10 ].

Intake recommendations for biotin and other nutrients are provided in the Dietary Reference Intakes DRIs developed by the Food and Nutrition Board FNB at the National Academies of Sciences, Engineering, and Medicine [ 1 ].

DRI is the general term for a set of reference values used for planning and assessing nutrient intakes of healthy people. These values, which vary by age and sex, include the following:. The FNB found the available data to be insufficient to derive an EAR and RDA for biotin.

For this reason, the FNB established only AIs for biotin. The FNB based its determination of AIs for all populations on the amount of biotin in human milk consumed by infants and then used body weight to extrapolate AIs for other groups [ 11 ].

Table 1 lists the current AIs for biotin [ 1 ]. Many foods contain some biotin. Foods that contain the most biotin include organ meats, eggs, fish, meat, seeds, nuts, and certain vegetables such as sweet potatoes [ 2 , 12 ].

The biotin content of food can vary; for example, plant variety and season can affect the biotin content of cereal grains, and certain processing techniques e. Cooking denatures avidin, making it unable to interfere with biotin absorption [ 13 ].

The U. Food and Drug Administration FDA developed DVs to help consumers compare the nutrient contents of foods and dietary supplements within the context of a total diet. The DV for biotin is 30 mcg for adults and children age 4 years and older [ 15 ].

FDA does not require food labels to list biotin content unless biotin has been added to the food. Biotin deficiency is rare [ 12 , 18 ], and severe biotin deficiency in healthy individuals eating a normal mixed diet has never been reported [ 13 ].

The signs and symptoms of biotin deficiency typically appear gradually and can include thinning hair with progression to loss of all hair on the body; scaly, red rash around body openings eyes, nose, mouth, and perineum ; conjunctivitis; ketolactic acidosis which occurs when lactate production exceeds lactate clearance and aciduria abnormal amounts of acid in urine ; seizures; skin infection; brittle nails; neurological findings e.

The rash and unusual distribution of facial fat in people with biotin deficiency is known as biotin deficiency facies [ 1 , 13 ]. Biotinidase deficiency is a rare autosomal recessive disorder that prevents the body from releasing free biotin, leading to biotin deficiency despite normal intake.

However, biotin interferes with lab work, resulting in false readings. This effect has been noted in the following tests:. Biotin use has caused the misdiagnosis of hyperthyroidism and Graves' disease , a serious autoimmune thyroid disease.

This is due to biotin's impact on thyroid lab work. Check with your healthcare provider before starting a supplement regimen. They may suggest that you stop taking biotin before getting your labs done. A daily adequate intake AI of 30 micrograms mcg of biotin is adequate for adults.

Although biotin is unlikely to cause adverse side effects, even in high doses, keep in mind that dietary supplements are not regulated the way drugs are in the United States.

This means the Food and Drug Administration FDA does not approve them for safety and effectiveness before products are marketed. When possible, choose a supplement tested by a trusted third party, such as the following:. Note that even if supplements are third-party tested, they are not necessarily safe for all or effective in general.

In all cases, patients receiving biotin supplementation had an underlying pathology for poor hair or nail growth. All cases showed evidence of clinical improvement after receiving biotin. In cases of acquired and inherited causes of biotin deficiency as well as pathologies, such as brittle nail syndrome or uncombable hair, biotin supplementation may be of benefit.

However, we propose these cases are uncommon and that there is lack of sufficient evidence for supplementation in healthy individuals.

No severe toxicities have been reported with biotin supplementation. Results may be falsely high or low. Incorrect test results may lead to inappropriate patient management or misdiagnosis.

For example, a falsely low result for troponin, a clinically important biomarker to aid in the diagnosis of heart attacks, may lead to a missed diagnosis and potentially serious clinical implications.

The FDA has received a report that one patient taking high levels of biotin died following falsely low troponin test results when a troponin test known to have biotin interference was used.

For dermatologists, thyroid function studies are the most likely tests where biotin interference will be observed. For example, dehydroepiandrosterone sulfate DHEASluteinizing hormone LHfollicle-stimulating hormone FSHtumor markers such as cancer antigen CACA and CA and troponine T.

The bit of thinning is very likely due to medications I take such as blood pressure meds. I have tried other mixed vitamins before that has biotin, and what felt like no results. I also know because I have to shave my legs more often again. Now this may only be me but this specific biotin I take 1x daily in the morning and with the same brand I take 1x daily vitamin D.

My hair growth sped up for sure. I have normal shedding in the shower. I have 2 of them so one I have hooked up in my shower and one in a drawer. I use the one in the shower to gently brush conditioner through my hair.

I only use conditioner mid shaft to ends. I have to wash my hair and scalp very throughly until I see no suds at the drain. If conditioner residue is on my scalp I tend to sweat in sleep. I use all of their scents. I have a ton of different brands but I keep going back to Hask. I cannot use a conditioner that I have a hard time brushing thorough and that brand works great for me.

I use usually shampoos and conditioners that have biotin and caffeine in them with collagen. I started losing my hair due to a major illness I had a couple years ago.

Without treatment, biotinidase deficiency produces neurological and cutaneous symptoms, and profound biotinidase deficiency can lead to coma or death [ 19 , 20 ]. Chronic exposure to alcohol inhibits the absorption of biotin [ 21 ]. At least a third of pregnant women develop marginal biotin deficiency in spite of normal biotin intakes; plasma and breastmilk concentrations of biotin decrease in lactating women, even when their dietary biotin intakes exceed the AI [ 2 , 18 , 22 ].

Additional research is needed to understand the clinical significance of these findings. Signs of biotin deficiency include skin rashes, hair loss, and brittle nails [ 10 , 13 ]. Therefore, biotin supplements are often promoted for hair, skin, and nail health [ 16 , 23 , 24 ].

However, these claims are supported, at best, by only a few case reports and small studies. The evidence on biotin supplementation to treat brittle nails includes three small studies that did not include a placebo group, and these reports do not indicate the baseline biotin status of study participants.

One of these studies assessed the effects of 2. In the second study, 2. Finally, the third, retrospective study in 35 patients with brittle nails found that 2.

Only case reports are available to support claims that biotin supplements can promote hair health, and these reports were only in children [ 28 , 29 ]. Future studies are needed to determine whether biotin supplements might improve hair, nail, and skin health, especially among healthy individuals.

The FNB was unable to establish ULs for biotin because there is no evidence in humans that biotin is toxic at high intakes [ 1 ]. High biotin intakes, and potentially even intakes greater than the AI, may pose another type of health risk [ 32 ].

Supplementing with biotin beyond recommended intakes can cause clinically significant falsely high or falsely low laboratory test results, depending on the test. These incorrect results may lead to inappropriate patient management or misdiagnosis of a medical condition. The following section has more details on these interactions.

Very high intakes of biotin may interfere with diagnostic assays that use biotin-streptavidin technology and are commonly used to measure levels of hormones such as thyroid hormone and other analytes such as hydroxyvitamin D, producing falsely normal or abnormal results [ 9 , 32 ].

Even a single 10 mg dose of biotin has interfered with thyroid function tests administered within 24 hours of taking the supplement [ 38 ].
